什么时do，什么时候用have，什么时候用is？

用am/is/are 提问：
1、一般现在时态中，主语＋be＋名词、代词、介词短语的时候；
如：Is
it
in the classroom?
Are these kites?
Are you Anne?
2、在现在进行时态中
如：Are you listening to the music?
3、一般现在时态的被动语态中
如：Is he taught to do so?

用do/does 提问：
一般现在时态中，谓语动词是行为动词的时候
如楼主的那句话便是一个很好的例子:
Do you speak English?
这里当然包括have表示“拥有”的情况
如：Do you have a dictionary?

用have/has 提问：
1、某人拥有某物时（have这个单词较特殊，两种方式都是对的）
如：Have you a new bike?
2、现在完成时态中
如：Have you finished your homework?
Have you had your lunch?

is 是 be 的一种形式.常作联系动词,后面接形容词、名词、介词等作表语.如：
He is a teacher.
She is beautiful.
Tom is behind the tree.
do 可以作为助动词,和主要动词一起连用,构成否定、疑问等.如：
I like music. -- I don't like music. Do you like music?
do 还可以作为实义动词,“做”的意思,用法跟其他实义动词一样.

系动词be用is(am,are,was,were); “有”动词用have(has,had); 其他实意动词用do(does, did)。
He is a doctor. They have lots of money. We do morning exercises every day.
Is he a nurse? Have they any money? Do they work eight hours a day?
He is not a worker. They have not any money. They do not know it.

问句的话
一般现在时的时候用do，一般完成时的时候用have，现在进行时的时候用is
